A common schedule is 0.25 → 0.5 → 1.0 → 1.7 → 2.4 mg weekly, holding each step about four weeks. The starting 0.25 mg is generally a tolerance-building dose.
Gradual escalation reduces gastrointestinal side effects — nausea is the most common — by giving the body time to adjust before each increase.
Around four weeks per step is typical, but the hold can be extended if a step isn't well tolerated.
Semaglutide vials are reconstituted with bacteriostatic water, and your dose is drawn on a U-100 insulin syringe marked in units.
Concentration (mg/mL) = vial mg ÷ water mL. Units = (dose mg ÷ concentration) × 100.
Enter your vial size and water volume in the reconstitution calculator to convert every mg step into exact units for your vial.
